PERBANDINGAN EFEKTIVITAS FIKSASI MENGGUNAKAN MADU DAN NBF 10% PADA KUALITAS PEWARNAAN HEMATOKSILIN-EOSIN: TINJAUAN LITERATUR SISTEMATIS

Neutral Buffered Formalin (NBF) 10% adalah standar utama fiksasi jaringan untuk pewarnaan Hematoksilin-Eosin, namun bersifat karsinogenik dan toksik. Madu berpotensi menjadi alternatif alami yang aman dan murah karena kandungan bioaktif serta sifat asamnya mampu mempertahankan morfologi jaringan secara sebanding. Karena hasil penelitian terdahulu masih bervariasi dalam konsentrasi, waktu, dan jenis jaringan, Studi Literatur Sistematis ini bertujuan untuk mengidentifikasi serta mengevaluasi efektivitas madu dibandingkan NBF 10% sebagai agen fiksatif secara komprehensif. Penelitian Studi Literatur Sistematis menggunakan model PRISMA untuk memperoleh artikel penelitian dari tahun 2016–2026 dari Google Scholar dan ScienceDirect yang diseleksi berbasis kriteria inklusi-eksklusi spesifik. Data yang terkumpul kemudian dianalisis secara deskriptif-komparatif guna memberikan gambaran komprehensif terkait pengaruh konsentrasi madu terhadap kualitas pewarnaan H&E sediaan histologis. Hasil menunjukkan bahwa Madu berpotensi sebagai fiksatif alternatif alami pengganti NBF 10% dengan pewarnaan Hematoksilin-Eosin karena sifat antimikroba, antioksidan, dan kemampuannya membentuk ikatan silang protein. Secara mikroskopis, madu mampu menghasilkan pewarnaan inti sel yang baik mendekati NBF 10%. Namun, kualitas pewarnaan sitoplasma dan preservasi makroskopisnya bervariasi karena jaringan cenderung lebih lembek. Efektivitas madu sangat bergantung pada konsentrasi, durasi fiksasi, dan jenis jaringan. Konsentrasi rendah berisiko memicu lisis sel, sedangkan konsentrasi terlalu tinggi dapat menyebabkan dehidrasi jaringan. Oleh sebab itu, diperlukan penelitian eksperimental lebih lanjut dengan protokol yang terstandarisasi untuk memperoleh bukti efektivitas madu sebagai alternatif NBF 10%.
Neutral Buffered Formalin (NBF) 10% is the primary standard for tissue fixation in Hematoxylin-Eosin staining, but it is carcinogenic and toxic. Honey has the potential to be a safe and inexpensive natural alternative because its bioactive compounds and acidic properties can preserve tissue morphology comparably well. Since previous research results have varied in terms of concentration, duration, and tissue type, this Systematic Literature Review aims to comprehensively identify and evaluate the effectiveness of honey compared to 10% NBF as a fixative. This Systematic Literature Review used the PRISMA guidelines to identify research articles published between 2016 and 2026 from Google Scholar and ScienceDirect, selected based on specific inclusion and exclusion criteria. The collected data were then analyzed using descriptive and comparative methods to provide a comprehensive overview of the effect of honey concentration on the quality of H&E staining in histological specimens. The results indicate that honey has the potential to serve as a natural alternative fixative to 10% NBF in Hematoxylin-Eosin staining due to its antimicrobial and antioxidant properties, as well as its ability to form protein cross-links. Microscopically, honey produced good nuclear staining comparable to 10% NBF. However, the quality of cytoplasmic staining and macroscopic preservation varied because the tissue tended to be softer. The effectiveness of honey is highly dependent on concentration, fixation duration, and tissue type. Low concentrations have the risk of causing cell lysis, while concentrations that are too high can cause tissue dehydration. Therefore, further experimental research using standardized protocols is needed to establish evidence of honey’s effectiveness as an alternative to 10% NBF.
